Ei-ichi Negishi

I still would like to go through my Nobel Prize-winning chemistry a little bit, mainly to provide what I would call ABC guidelines for young people. So, I urge young people to follow, as I do, what I call ABC guidelines. A word is ambitious, and people ask me, how ambitious should I be, should we be. Well, of course, as ambitious as can be. And the B word, two words I propose, be basic, you know, pay attention to the basic, and in a broad sense, as broadly approach, the broader the better. And the C word, of course we all must be, in this business, creative, be creative. I think that's what I understand the Nobel Foundation pay most maximum attention to. But I also have the second keyword, which I have been focusing my attention. That's catalysis, and many, many things, as we have just heard, biochemistry and here and there, things are catalytic, should be catalytic. And that's what, with those in mind, let me just spend several minutes, okay, so I think we can, we can, oops, how to advance, how to advance the slide. Yeah, here? So here it is, this is the entire universe, as far as chemists are concerned, as you know. And here you see 113 or so elements, and Mendeleev, way back one and a half century ago, he already aligned them, about 90 of them, 90. That is almost 80 percent of the universe. And we cannot, right now, go out of this territory, so we must stay, more or less. But let me quickly tell you, as you already know, okay, so this is No. 84. From 84 onward, most of them are radioactive, and for organic synthesis, it's very difficult to deal with them. So we, by and large, avoid them. There's also technetium here. Okay, and then, of course, inert gas, useful but they don't enter into organic synthesis so much. They support us. And then there are close to 10 intrinsically toxic ones: beryllium, cadmium, you know, this cadmium poisoning is very bad. And all Japanese know mercury poisoning, Minamata disease. Thallium, lead, tin or so. Tin has been used, somewhat unfortunately, widely in our cross-coupling. But tin is toxic, so we probably should stay away from it. And then, of course, this toxic milk. So when you eliminate all these and all these, then we're still left with about 70 elements. So the universe provides us about 10 or so what I call organic elements. And then this blue-ish, 20s, a little more than 20, main group metals. They are relatively inexpensive, so we may use them in a stoichiometric way, where, of course, catalytic ones are better. But my attention has been focused, over the past 40 or so years, in this area. Three times eight, 24, minus one technetium, 23 golden, it's not yellow, it's golden, golden-coloured d-block transition metals. They are wonderful catalyst sources. Many of them can participate in the wonderful catalysis. And in my opinion, the more we use them in a highly catalytic way, I think the future will be brighter in the 21st century. Anyway, that's what I have been paying my attention on. And let's see, so here, if you visit Saint Petersburg and Moskovsky Avenue and there's this Mendeleev Institute, so Mendeleev here. And he already knew, or placed correctly, about 90 elements, as I told you, and of about 90, he was credited, you know, for the discovery of 11 of them. Of course, Madame Curie, received two Nobel Prizes for discovering, not just for discovering but, two new elements. Anyway, he came half a century too early, so he got zero. But to me (laughs), (audience laughter) to me, he's the founding father of the modern chemistry. Oops, all right, yeah. Okay, and so, yeah, okay. So getting back to my ABC guideline, how should we synthesize compounds, or how should we synthesize organic compounds, in my case. Well, I believe that in a Y-E-S-E-S way, I got this catchphrase from Barack Obama, you know, several years ago when he was saying yes, yes, yes, yes. So I wanted to say yes-yes way, but I couldn't find, I have never found a second Y word, so mine is a Y-E-S-E-S, okay. What is Y? Well, of course, in any synthesis, your yield should be as high as, as close to hundred percent as possible, the yield. And the E word, my first E word is efficiency. We want to synthesize any compounds in as few steps as possible. S, selectivity should be very high, preferably 6/9 or 7/9. My friends in pharmaceutical chemistry would say, well, 6/9s are usually okay, but sometimes it's borderline. If you can synthesize organic target molecules in 7/9 purities, then even a minute amount of toxic material may not serve, so I'm not sure. So that's the borderline level that we try to achieve or go over. So selectivity is very important. As I said, I have not found a second Y word. Please, if you can come up with this, I don't know, I'm not rich, but I will deeply thank you. And a second E word is, anyway, so let me just go, okay. And the second E word is efficiency, the step-economy. Did I say that? And the second S word is step-economy. Sorry, the second S word is safety. Safety, which I already told you. And then my Nobel Prize-winning work goes like this. Before us, to synthesize various organic compounds, about 10 classes here with the metals and 10 classes or so here, or 8 or 9, I forgot, some X halogen and electronegative groups. And then you have about close to 10 by 10 matrix. And then if you can synthesize all of them, then you might be in reasonably good shape. When I was 26 as a PhD student at the University of Pennsylvania, I came up with this LEGO game approach, which now is called the cross-coupling. I used to call this the LEGO game, because in the LEGO game, LEGO game was sweeping, invading America, from Denmark. And as you know, there are many pieces with a hole, so that's like organic halide that is electron deficient. And then there are other many pieces, as many as you can think of and prepare, with a stick. So here, with the two electrons, this represents organic metals. So you make all kinds of them, any kinds you want, and then bring them and snap them. Then you can synthesize all kinds of organic compounds, so I thought. But and, of course, we learned the Grignard chemistry with Mg here. And I went to the library, extensively checked and summarized as shown here. Here without catalyst, virtually nothing works well. Outside this, they sometimes work, but there are selectivity issues and others, you're in trouble. Okay, so here I thought maybe we should, fundamentally improve this situation. And we started, when I became assistant professor, 1972, at the Syracuse University, I started. And then within several years, we were able to paint this scheme in this way. So going from this to this, so some of my friends said: "Wow, that's a miracle." There's no miracle in chemistry, perhaps, but it has been substantially improved, as you can see. There are some trouble areas, but these products are obtainable through these processes. You can also obtain this very nicely. So we usually don't need this, but anyway, so it's not perfect. But this is the scheme the Nobel Foundation kindly recognized. Okay, but that'... Why do we want to use metals? Very, very simple, basic question. Well, if you mix alkane, saturated alkanes, with alkenes, there's no facile reaction, as you know. But if you convert this alkane into carbocations, then, of course, a very rapid wide reaction takes place. Significance of this empty orbital is vividly shown, indicated, here. No wonder my professor, Professor Brown's, hydroboration works so rapidly, even at minus 70 or 78 degrees. The reaction does proceed. And the secret here is, of course, a combination that's C-H bond, along with this empty orbital. And if you compare this with this, they are, sort of isoelectronic. And if you stretch your imagination, then even this one, hydrozirconation, with 16 electrons, I believe. And you know, this is very, what Roald Hoffmann might call isolobal. Isolobal situation, no wonder they are all so low. An important thing is that these reactions are strictly, or essentially, as far as we can tell, nearly hundred percent stereoselective syn addition. Okay, so here is a very, very educational, very useful summary that I learned a lot from. It was published rather recently, 10, 12 years ago, but this was probably known much earlier. That is that if you mix acid with base, with a pair of electrons, you might think that the acid-base reaction goes "bing", they will go almost spontaneous way under all kinds of conditions. But they analyse this one in a theoretical chemical way, and they said there is a big barrier to this reaction, actually, a significant barrier. I think this is very important. As B begins to donate a pair of electrons to A, the A becomes negatively polarized, and the B becomes positively polarized. What is the effect of this polarization? Well, this negative charge, you will begin to say, these incoming electrons go back. Electrons are negative, negative and negative and they their repell each other. And at base here, with a positive polarization, it starts saying "come back", because these electrons are negative. So this was indicated here. And so the key message is that a one-way two-electron transfer intrinsically involves a high energy barrier. How do we solve that? We'll make it two ways, which was suggested first, in my opinion, by Fukui. He was doing this sort of thing during World War II, early part of 1940s, but he didn't have a chance to report. Japan at that time was in a terrible situation. And Fukui was the one who actually introduced the concept of LUMO, you know, we shouldn't be just focusing our attention on the HOMO. There's always a LUMO, empty LUMO. And then they play a very, very key, important, well, I'm not trying to combat Fukui's theory, but I'm just trying to show. So and then, but he published his paper in '51 or so. And it was in a year or two in the UK, M. J. S. Dewar proposed this so-called famous Dewar-Chatt-Duncanson model. So here we have a transition metal or species, metal species with a block HOMO and then LUMO. So d-block transition metals can really provide this kind of HOMO-LUMO combination species. That is a very, very important recognition. But along with Fukui's alkene, alkenes we normally think of the sigma-bond and the pi-bond. But he says that's not enough, we must think of, always think of the one above this pi-bond that is empty, but that's a LUMO. And he told us that this HOMO-LUMO and HOMO-LUMO interaction, these two interactions are fundamentally critically important. So in one case inside two electrons go from bottom to up, bottom-up, notice this direction, and then outside from top to bottom. Why is this movement important? Let's go back to this one. You see, a one-way two-electron interaction has a high energy barrier. If it can be made two ways, this way, then those ill effects can be pretty much cancelled. So this is the essence of why we should be using d-block transition metals as a catalyst, because they can form species that can be represented by something like this. So they can really participate in two-way four-electron transfer reactions. And this, to me, explains the majority, if not all, many, many of fine transition metal catalysed reactions. Okay, but there are some other ways. Hydrometalation, Brown's hydrometalation. Why is this so fast and almost hundred percent syn addition? Well, if you accept Fukui's explanation, then we can write the same HOMO-LUMO, and then HOMO, HB, and then a LUMO, so six electron species, so two up, two down. And hundred percent syn addition reaction. Yields are usually very, very high. Now what I, as an independent, young assistant, not so young, assistant professor, I said, by looking at this thing, Brown's, then I realized that we can, almost at will, change this part, supposedly change this part, from hydrogen to all kinds of metals in the periodic table. For instance, changing this to carbon, two up, two down. I even named this one, before I discovered my first one, carbometalation. And when we came up with this carbometalation, I used this term carbometalation. I was scolded by many senior colleagues: "What are you doing, what is this carbometalation, what does it mean?" You don't want to mess up a technical term, jargon, or whatever. But I said, well, if this is hydrometalation, shouldn't we call this carbometalation. I beared with it, and in maybe 10 years' time, many of them started using this term, carbometalation. Anyway, so I realized, but maybe this is the only thing that I can talk today, but maybe, so be it. But if you accept this hydrometalation and the carbometalation, then you realize that you can put all kinds of metals here to come up with metalometalation, and today there are many of them known. Now, see halometalation or heterometalation, that covers the whole periodic table. That's what I want to do, you know, I want to play with this, Mendeleev presented the periodic table. Whatever I do, I want to go all over. Okay, so let me ask this question. Some young people might have immediate answers. With heterometalation, this is like halogen or oxy or, you know, those electronegative metals, such reactions are very rare. You know why, why, anyone, anyone? No, anyway, of course, with the heteroatoms here, this bond, the M-X bond, is strong, then you have to break it. Could be contra-thermodynamic in many cases. But turned out that already in 1964 in the UK, Mike Lappert, I think that's his name, I'm not sure about the correctness of the name. He already published a haloboration. So boron is a metal, well, boron is almost not metal, as some people might say. This haloboration was already published in 1964, and then, as you might guess, halogen and boron at hundred percent syn manner. Okay, I think, so in this way we discovered cross-coupling reactions. And once they are discovered, you can synthesize all kinds of molecules, not just the kinds that you want to synthesize, in a very highly selective way. Let me just show you 8-trienol, 8-ester. Or trans 1, 2, 3, 4, these had been synthesized before us. But look at this uniformly high yield and uniformly high purity. So okay, some of these, with much difficulties, synthesized before us, but none of the other four. There are, you know, 2 times 2 times 2, 8 stereoisomers, okay. So this one, no one had synthesized before us. With this cis bond in here it was a very difficult proposition. But now if you do it in a coupling way, there's no problem. So in very high excellent yield, in nearly hundred percent stereoselectivity to finish, complete, these two are, even in our hand, somewhat difficult. We needed Mike Organ's PEPPSI catalyst. Don't ask me what the structure is. I may not be able to draw today. But in excellent yield and the nearly hundred percent stereoselectivity, published only a few, several years ago. Okay, but now, once you can do that, well, CoQ10, you know, and these things, once you synthesize very simple pieces, in less than 10 steps we can synthesize them. And actually, in Japan there's the Kaneka. They are not using this method yet, but they are very much interested in it. Okay, actually, I meant to present this kind of, no double bond, no aromatic, no triple bond kind of molecules, in an asymmetric way. But I would be crazy if I start, I won't do that. But for these reactions, I will just show you that we needed to discover, as I said, ABC, discover what we will call ZACA reactions, zirconium-catalysed asymmetric carbo-alumination of alkenes. This is a one-step Ziegler-Natta reaction, one-step. And okay, but there are many differences. In the Ziegler-Natta reaction, people usually do not care, hardly, about the absolute stereochemistry, for instance, centre. They are concerned mainly about tacticity. If one method goes this way, next one should also be this way and this way. And that's isotactic. But if you make a polymer and if you just turn this around, then that could be an enantiomer. So absolute stereochemistry doesn't matter. Here absolute stereochemistry of course does matter. And it was a very difficult process, took me, took us, 17 long years, because this is what we wanted to do but all these things are blocking our way. But we managed to avoid them. And now, once we synthesize this, this one my friend, Kenji Mori, made this thing, 20-some steps, 20 steps. Now we can make it 10 steps and so on. And so just to go to... So we are very, very proud of this chemistry, and I will go all the way, look at this thing, for instance. We control this chirality here at this centre, and this side is CH3-CH2. This side is CD3-CH2. We call them isotopomers. So once you come to this region, then you can maybe... Yeah, we were sure that we made them, but how we didn't know, we ran into a problem, how to determine, analytically determine, isotopomers. Okay, so that was, the isotopomer was branching in the beta position, but now, more recently, in this case, this is also isotopomer. This is a CH3-CH2, CD3-CH2, alpha, beta, gamma position. So this is more difficult, and alpha, you know, an optical rotation. Alpha D is too small to be determined. So we have come to the other dead end. This one is alpha, beta, gamma, delta, so this a more difficult rotation, not detectable. Finally, this one, alpha, beta, gamma, so sorry, yeah, okay, anyway. These things are ND, means nondetectable. So what... Alpha, beta, gamma, delta, epsilon isotopomer. We run out of the chromatographic tools, as far as we are concerned. The only thing that survived, luckily, was NMR, yeah, NMR. And so first we were successful with a Mosher ester analysis for analysing this thing. This is not isotopomer yet, but they're very remote. And eventually, we needed to shift from Mosher ester to Tokyo Institute of Technology guys. This one has a longer reach, and as you can see, diastereomeric mixture, you can see two set, the sets of doublets, you know, the here, here, here. And when we synthesize, we see something like this. We see, we don't see this, we see this, we don't see this, we see this, we don't see this. It's pure, so our method can synthesize, you know, give us isomerically, very, very pure, isotopomeric, or whatever you call. And we now claim that we have reached, you know, we have conquered one of the last bastions in organic synthesis, that's the frame that I'm using. And then not only that, very, very, molecule like this, they are very important, tuberculosis vaccine and so on. And thess components you can see here and there. And this was synthesized first in the Netherlands, 27 steps, 25, you know, you can just see this. It was the pioneering original synthesis. And it was shortened to 20, and overall yield went up to 15. You calculate the average yield of each step, they must maintain over 90% throughout. Ours is very, very short. Here we still lack significant, somewhat, I should say. So in our case, we synthesize this by ZACA reaction, and this or these, and then you just snap them, by cross-coupling, to here, and then we have this, eight longest linear steps. Okay, so that's the end of my story, and over a hundred people have participated in the cross-coupling discovery and development. And in the case of ZACA chemistry, it was actually started early with alkynes, but I'm highlighting some of the key players and the recent players. And this is my recent group, small group. Ich möchte noch etwas bei meiner Nobelpreis-prämierten Chemie bleiben, vor allem, um jungen Menschen die sogenannten ABC-Richtlinien darzulegen. Ich ermuntere die jungen Menschen dazu, wie ich diese ABC-Richtlinien zu befolgen. Das Wort mit A ist ambitious (ehrgeizig) und man fragt mich, wie ehrgeizig man bzw. wir sein sollten. Natürlich so ehrgeizig wie möglich. Und das B-Wort, zwei Worte, die ich vorschlage, halten Sie es basic (einfach), achten Sie auf Grundlegendes und auf einen broad sense (einen breiteren Ansatz), je breiter umso besser. Und das C-Wort, wir alle sollten natürlich creative (kreativ) bei dieser Sache sein, seien Sie kreativ. Dem schenkt, wie ich denke, die Nobel-Stiftung die maximale Aufmerksamkeit. Ich habe aber auch ein zweites Schlüsselwort, das ich befolge. Das heißt Katalyse und viele Sachen, wie wir gerade gehört haben, Biochemie und so weiter, Dinge sind katalytisch, sollten katalytisch sein. Und das ist, mit dem Gedanken daran, lassen Sie mich nur einige Minuten sprechen, ich denke, wir können, wir können... Wie geht das? Wie kann man die Folie wechseln? Ja, hier? Also hier ist es, das ganze Universum, zumindest was die Chemiker angeht, wie Sie wissen. Und hier sehen Sie 113 oder mehr Elemente und Mendelejew, lange ist das her, ein halbes Jahrhundert, er hatte sie bereits angeordnet, etwa 90 von ihnen, 90. Das sind fast 80 Prozent des Universums. Und wir können uns nicht, nicht jetzt, aus diesem Gebiet entfernen, wir müssen bleiben, mehr oder weniger. Aber lassen Sie mich schnell sagen, wie Sie bereits wissen, OK, das ist die Nr. 84. Und alle Japaner kennen eine Quecksilbervergiftung, die Minamata-Krankheit. Thallium, Blei, Zinn oder so. Zinn ist leider, unglücklicherweise, bei unserer Kreuzkupplung weit verbreitet. Aber Zinn ist giftig, also sollten wir uns wohl besser von ihm fernhalten. Und dann gibt es natürlich diese giftige Milch. Also, wenn Sie all diese eliminieren, dann gibt es immer noch ca. 70 Elemente. Das Universum stellt uns etwa 10 bereit, die wir organische Elemente nennen. Und dann diese Bläulichen, 20, etwas mehr als 20, Metalle der Hauptgruppe. Sie sind relativ billig, so dass wir sie in einer stöchiometrischen Weise verwenden können, aber die Katalytischen sind natürlich besser. Aber ich habe meine Aufmerksamkeit in den letzten 40 Jahren auf dieses Gebiet gerichtet. Dreimal acht, 24, minus einem Technetium, 23, golden, es ist nicht gelb, sondern golden, goldfarbene D-Block-Übergangsmetalle. Es sind wunderbare Katalysatorquellen. Viele von ihnen können an der wundervollen Katalyse teilnehmen. Und ich denke, dass, je mehr wir sie in einer stark katalytischen Weise anwenden, umso besser wird die Zukunft des 21. Jahrhunderts sein. Wie auch immer, darauf habe ich meine Aufmerksamkeit gerichtet. Und lassen Sie uns schauen, hier, falls Sie Sankt Petersburg und die Moskovsky- Straße besuchen, gibt es dieses Mendelejew-Institut, Mendelejew ist hier. Und er wusste oder hatte bereits etwa 90 Elemente, wie zuvor erwähnt, richtig eingeordnet und von etwa 90 wurden ihm, wissen Sie, die Entdeckung von 11 von ihnen zugeschrieben. Madame Curie erhielt natürlich zwei Nobelpreise für die Entdeckung, nicht nur für die Entdeckung, sondern zwei neue Elemente. Er war ein halbes Jahrhundert zu früh dran und erhielt gar nichts. Aber für mich (lacht), (Gelächter im Publikum) für mich ist er der Begründer der modernen Chemie. Huch, alles OK, ja. OK. Ja, OK. Also zurück zu meiner ABC-Richtlinie. Wie sollen wir Verbindungen synthetisieren oder wie sollten wir, in meinem Fall, organische Verbindungen synthetisieren? Nun, ich denke, auf eine „J-A-A-A-Art und Weise‘‘, diesen Spruch habe ich von Barack Obama, als er vor einigen Jahren ja, ja, ja, ja sagte. Ich wollte es den ja-ja Weg taufen, habe aber nie ein zweites J-Wort gefunden, also ist meines J-A-A-A. Was ist J? Nun, natürlich sollte in jeder Synthese das Ergebnis so exakt wie möglich sein, so nahe an hundert Prozent liegen wie möglich, der Ertrag, Und mein E-Wort, meine erstes E-Wort heißt Effizienz. Wir möchten jeden Komponenten in so wenigen Schritten wie möglich synthetisieren. S, die Selektivität, sollte sehr hoch sein, vorzugsweise bei 6/9 oder 7/9 liegen. Meine Freunde aus der pharmazeutischen Chemie würde sagen: 6/9 sind in der Regel in Ordnung, aber manchmal sind sie nicht ausreichend. Wenn Sie organische Zielmoleküle in 7/9-Reinheiten synthetisieren, dann braucht man vielleicht nicht einmal eine geringe Menge an toxischen Material, also ich bin mir nicht sicher. Das ist also die Grenzebene, die wir versuchen zu erreichen oder zu überschreiten. Die Selektivität ist also sehr wichtig. Wie ich schon sagte, habe ich kein zweites J-Wort gefunden. Bitte, falls Ihnen eines einfällt, ich bin nicht reich, aber ich werde Ihnen überschwänglich danken. Und ein zweites E-Wort, wie auch immer, lassen Sie mich einfach weitermachen. Das zweite E-Wort heißt Effizienz, Schritt-Wirtschaft. Hatte ich das gesagt? Und das zweite S-Wort ist Schritt-Wirtschaft. Entschuldigung, das zweite S-Wort ist Sicherheit. Sicherheit, wie ich schon gesagt hatte. Und meine mit dem Nobelpreis ausgezeichnete Arbeit lautet so: Vor uns, um verschiedene organische Verbindungen zu synthetisieren, etwa 10 Klassen hier mit Metallen und 10 Klassen oder dort, oder 8 oder 9, ich weiß nicht mehr, einige X-Halogene und elektronegative Gruppen. Und dann sind Sie nahe an einer 10 mal 10-Matrix. Und dann, wenn Sie alle synthetisieren können, dann sind Sie gut mit dabei. Als ich ein 26 jähriger Doktorand der Universität von Pennsylvania war, erfand ich diesen LEGO-Spiel-Ansatz, der jetzt als Kreuzkopplung bezeichnet wird. Ich nannte es das LEGO-Spiel, denn beim LEGO-Spiel, das LEGO-Spiel hatte Amerika überrannt, kam aus Dänemark. Und wie Sie wissen gibt es viele Teile mit einem Loch, wie ein organischer Halogenid mit Elektronenmangel. Und dann gibt es noch viele andere Teile, so viele, wie Sie sich vorstellen können, die man zusammenstecken kann. Hier, mit den beiden Elektronen, das stellt organische Metalle dar. Sie können alle möglichen Konstruktionen kreieren und sie dann zusammenstecken. Dann können Sie also alle Arten von organischen Verbindungen synthetisieren, dachte ich. Aber natürlich kennen wir die Grignard-Chemie mit Mg hier. Und ich ging in die Bibliothek und habe das Ganze umfangreich geprüft und wie hier dargestellt, zusammengefasst. Hier arbeitet ohne den Katalysator praktisch gar nichts gut. Außerhalb arbeiten sie manchmal, aber es gibt Selektivitätsprobleme und Sonstiges, man ist in Schwierigkeiten. Okay, an dem Punkt dachte ich, vielleicht sollten wir diese Situation grundsätzlich verbessern. Und wir begannen damit als ich 1972 Assistenzprofessor an der Syracuse University wurde. Und dann, innerhalb von einigen Jahren ist es uns gelungen, dieses Schema auf diese Weise zusammenzustellen. Also von dem hier zu dem, und meine Freunde sagten: „Wow, das ist ein Wunder." Es gibt keine Wunder in der Chemie, eher nicht, aber es hat sich Einiges deutlich verbessert, wie Sie sehen können. Es gibt einige Problembereiche, aber diese Produkte kann man durch diese Prozesse erhalten. Sehr schön kann man das erhalten. Wir brauchen das in der Regel nicht, aber egal, es ist also nicht perfekt. Aber das ist das Schema, was die Nobel-Stiftung freundlicherweise anerkannt hat. Okay, aber das ist ... Aber wenn Sie dieses Alkan in Carbokationen konvertieren, dann, natürlich, findet eine sehr schnelle und umfangreiche Reaktion statt. Die Bedeutung dieses leeren Orbitals wird anschaulich gezeigt, hier dargestellt. Kein Wunder, dass die Hydroborierung von Professor Browns so schnell arbeitet, selbst bei minus 70 oder minus 78 Grad. Die Reaktion geht weiter. Und das Geheimnis hier ist eine Kombination, die CH-Bindung, zusammen mit diesem leeren Orbital. Und wenn Sie dies mit dem vergleichen, sind sie, irgendwie isoelektronisch. Und wenn Sie Ihre Vorstellungskraft erweitern, dann sogar diese hier, Hydrozirkonierung, mit 16 Elektronen, glaube ich. Und wissen Sie, das ist es, was Roald Hoffmann als isolobal bezeichnen könnte. eine Isolobal-Situation, kein Wunder, sie sind alle sehr niedrig. Eine wichtige Sache ist, dass diese Reaktionen streng oder wesentlich sind, soweit wir das beurteilen können, eine fast hundertprozentige stereoselektive Syn-Ergänzung. OK, hier gibt es eine sehr lehrreiche, sehr nützliche Zusammenfassung, von der ich viel gelernt habe. Sie wurde eher kürzlich, vor 10, 12 Jahren veröffentlicht, aber war wahrscheinlich schon viel früher bekannt gewesen. Wenn Sie Säure mit Base mischen, mit einem Paar Elektronen, könnte man denken, dass die Säure-Base-Reaktion ‘‘Bing" macht, unter allen Bedingungen ihrem fast spontan Weg folgt. Aber sie analysierten das auf theoretische, chemische Weise und sagten, dass es ein großes Hindernis bei dieser Reaktion, eine signifikante Barriere gibt. Ich denke, dass das sehr wichtig ist. Wenn B beginnt, ein Elektronenpaar an A abzugeben, wird A negativ polarisiert und B wird positiv polarisiert. Was ist die Wirkung dieser Polarisation? Nun, diese negative Ladung, werden Sie sofort sagen, diese ankommenden Elektronen gehen zurück. Elektronen sind negativ, negativ und sie stoßen sich gegenseitig ab. Und an der Basis hier, mit einer positiven Polarisation, beginnt es zu sagen „komm zurück", weil diese Elektronen negativ sind. Das war hier angegeben. Die Kernaussage ist also, dass ein Ein-Weg-Zweielektronen-Transfer eine hohe Energiebarriere bedeutet. Wie lösen wir das? Und dann spielen sie eine sehr, sehr wichtige Rolle, gut, ich versuche nicht, Fukuis Theorie zunichte zu machen, ich versuche, es zu veranschaulichen. Und dann veröffentlichte er seine Entdeckung um 1951 herum. Und es war 1 oder 2 Jahre zuvor in Großbritannien, dass MJS Dewar das so genannte berühmte Dewar-Chatt-Duncanson-Modell vorgestellt hatte. Hier haben wir also ein Übergangsmetall oder Metallarten mit einer Block-HOMO und LUMO. D-Block-Übergangsmetalle können wirklich diese Art von HOMO-LUMO- Kombinationsarten liefern. Das ist ein sehr, sehr wichtiger Ansatz. Aber zusammen mit Fukuis Alkenen, Alkene erinnern uns in der Regel an die Sigma-oder Pi-Bindung. Aber er sagt, dass das nicht genug sei, das wir immer an das über der Pi-Bindung denken müssten, dass ist leer, aber es ist ein LUMO. Und er hat uns gesagt, dass diese HOMO-LUMO und HOMO-LUMO-Wechselwirkung, diese beiden Wechselwirkungen von entscheidender Bedeutung sind. In einem Fall bewegen sich im Inneren zwei Elektronen von unten nach oben, achten Sie auf die Richtung, und dann außen von oben nach unten. Warum ist diese Bewegung so entscheidend? Gehen wir zurück zu diesem hier. Wie Sie sehen, eine Ein-Weg-Zwei-Elektronen-Wechselwirkung hat eine hohe Energieschwelle. Wenn es zwei Wege gibt, diesen Weg, dann können diese negativen Auswirkungen eliminiert werden. Das ist der Grund, warum wir D-Block-Übergangsmetalle als Katalysator verwenden sollten, denn sie können Arten bilden, die durch so etwas wie hier dargestellt werden können. So können tatsächlich an den Zwei-Wege-Vierelektronentransfer-Reaktionen teilnehmen. Und das erklärt mir die meisten, wenn nicht alle, viele, viele feine Übergangsmetall-katalysierte Reaktionen. Okay, aber es gibt noch andere Möglichkeiten. Hydrometallierung, Browns Hydrometallierung. Warum geht das so schnell und ist eine fast hundertprozentige Syn-Ergänzung? Nun, wenn Sie Fukuis Erklärung annehmen, dann können wir die gleiche HOMO-LUMO und dann eine HOMO, HB, und dann ein LUMO schreiben, also sechs Elektronenarten, zwei oben, zwei unten. Und eine hundertprozentige Syn-Ergänzungsreaktion. Die Erträge sind in der Regel sehr, sehr hoch. Nun, was ich als unabhängiger, junger Assistent, nicht so jung, Assistent des Professors sagte, als ich mir Browns Ding anschaute, dann bemerkte ich, dass wir beinahe nach Belieben diesen Teil abändern können, von Wasserstoff zu aller Art von Metallen, die dieses Periodensystem hat. Zum Beispiel, wenn wir das in Kohlenstoff umwandeln, zwei oben, zwei unten. Ich habe es sogar benannt, bevor ich meine erste entdeckte, die Carbometallierung. Wie auch immer, aber vielleicht ist dies die einzige Sache, die ich heute erzählen kann, aber vielleicht, lassen wir das. Aber wenn Sie diese Hydrometallierung und die Carbometallierung akzeptieren, dann werden Sie feststellen, dass man alle Arten von Metallen hier hinsetzen kann die mit einer Metalometalation kommen und heute sind viele von ihnen bekannt. Diese Haloboration wurde bereits im Jahr 1964 veröffentlicht und dann, wie Sie sich vorstellen können, Halogen und Bor bei hundertprozentiger Syn-Weise. OK, ich denke, auf diese Weise entdeckten wir die Kreuzkupplungsreaktionen. Und sobald sie entdeckt werden, können Sie alle Arten von Moleküle synthetisieren, nicht nur die, die Sie synthetisieren wollen, auf sehr hochselektive Art und Weise. Lassen Sie mich Ihnen 8-Trienol, 8-Ester zeigen. Oder trans-1, 2, 3, 4, diese wurden vor uns synthetisiert. Aber schauen Sie sich diesen gleichmäßig hohen Ertrag und die gleichbleibend hohe Reinheit an. OK, einige von ihnen synthetisierten mit viel Schwierigkeiten vor uns, aber keine der vier anderen. Es gibt, wissen Sie, 2 mal 2 mal 2, 8 Stereoisomere, OK. Also das hatte niemand vor uns jemals synthetisiert. Mit dieser Cis-Bindung hier war das es eine sehr schwierige Sache. Aber wenn Sie jetzt das Kupplungsverfahren anwenden, gibt es keine Probleme. Bei also einem sehr hohen ausgezeichneten Ertrag, fast hundertprozentiger Stereoselektivität das zu beenden, die beiden sind, sogar in unseren Händen, etwas Schwieriges. Wir brauchten Mike Organs PEPPSI-Katalysator. Fragen Sie mich nicht nach der Struktur. Ich kann sie heute vielleicht nicht zeichnen. Aber sie wurde mit ausgezeichnetem Ertrag und der fast hundertprozentigen Stereoselektivität erst vor ein paar Jahren veröffentlicht. OK, wenn Sie das erst einmal machen können, CoQ10, wissen Sie, und diese Sachen, wenn Sie sehr einfache Stücke synthetisieren, wir können sie in weniger als 10 Schritten synthetisieren. Und in Japan gibt es die Kaneka. Sie benutzen diese Methode noch nicht, aber sie interessieren sich sehr dafür. OK, eigentlich wollte ich diese Art vorstellen, keine Doppelbindung, keine Aromaten, keine Moleküle mit Dreifachbindung, in einer asymmetrischen Art und Weise. Aber ich wäre verrückt, wenn ich anfangen würde, das werde ich nicht tun. Aber ich werde Ihnen nur zeigen, dass wir für diese Reaktionen etwas entdecken mussten, wie gesagt, ABC, etwas, was wir ZACA Reaktionen nennen, Zirkonium-katalysierte, asymmetrische Carbo-Aluminierung von Alkenen. Dies ist eine einschrittige Ziegler-Natta-Reaktion, einschrittig. Und OK, aber es gibt viele Unterschiede. Bei der Ziegler-Natta-Reaktion kümmern sich die Leute kaum um die absolute Stereochemie, beispielsweise, Mitte. Sie achten hauptsächlich auf die Taktizität. Wenn eine Methode auf diese Weise funktioniert, sollte das die nächste auch usw. Und das ist isotaktisch. Aber wenn Sie ein Polymer kreieren und das umbiegen, dann könnte das ein Enantiomer sein. Absolute Stereochemie spielt also keine Rolle. Hier spielt die absolute Stereochemie natürlich eine Rolle. Und es war ein sehr schwieriger Prozess, der uns 17 Jahre lang beschäftigte, denn das wollten wir tun, aber alle diese Dinge blockieren den Weg. Aber wir haben es geschafft, sie zu vermeiden. Und nun, wenn wir sie nun synthetisieren, mein Freund Kenji Mori machte diese Sache, 20-oder so Schritte, 20 Schritte. Jetzt können wir es in 10 Schritte erledigen. Und so weiter... Wir sind also sehr stolz auf diese Chemie und ich werde durch das ganze Programm gehen, gucken Sie sich das hier an, zum Beispiel. Und wir hatten anfangs Erfolg mit der Mosher-Ester-Analyse. Das ist kein Isotopomer, noch nicht, aber sehr weit entfernt. Und schließlich mussten wir vom Mosher-Ester zum Institute der Technologie in Tokio umziehen. Es ist rein und unsere Methode kann synthetisieren, wissen Sie, isomerisch, sehr, sehr rein, isotopomerisch oder wie auch immer Sie es nennen. Und wir behaupten, dass wir es erreicht haben, wir eine der letzten Bastionen der organischen Synthese erobert haben, das ist der Rahmen, den ich verwende. Und dann, nicht nur das, sehr, sehr, Moleküle wie diese, sind sehr wichtig, ein Tuberkuloseimpfstoff usw. Und diese Komponenten können Sie hier und da finden. Und dies wurde zuerst in den Niederlanden synthetisiert, in 27, 25 Schritten, das können Sie hier sehen. Es war die wegweisende Original-Synthese. Und sie wurde auf 20 verkürzt und der Gesamtertrag stieg auf 15. Sie berechnen den durchschnittlichen Ertrag jedes einzelnen Schrittes, sie müssen mehr als 90% des Ganzen erzielen. Unserer ist sehr, sehr kurz. Hier fehlt noch etwas Bedeutendes, würde ich sagen. Also in unserem Fall synthetisieren wir das mit der ZACA-Reaktion und dem oder diesem und dann können Sie es durch die Kreuzkupplung verbinden und dann haben wir die hier, die acht längsten linearen Schritte. Okay, das ist das Ende meiner Geschichte, und mehr als hundert Personen haben an der Entdeckung und Entwicklung der Kreuzkupplung teilgenommen. Und im Fall der ZACA-Chemie hat es eigentlich früh mit Alkinen angefangen, aber ich möchte einige der wichtigsten Akteure und der jüngsten Spieler hervorheben. Abstract

The discovery and synthetic applications of a widely applicable and highly enantioselective (>99% ee) protocol consisting of the ZACA reaction (Zr-catalyzed asymmetric carboalumination of alkenes), purification of the ZACA-products by lipase-catalyzed acetylation, and subsequent transformations, such as Pd- or Cu-catalyzed cross-coupling are discussed.

In contrast with many other methods for asymmetric transformations requiring directing groups in the substrates, the ZACA–Lipase-catalyzed acetylation–Transition metal-catalyzed cross-coupling protocol in principle requires only simple alkenes. In practice, however, the presence of a proximal hetero-functional group can be tolerated for converting terminal alkenes into a wide range of chiral organic compounds including even those displaying very feeble optical rotations, such as isotopomers, as highly pure compounds of 99% ee or even higher. We now believe that one of the last bastions in organic synthesis may have been conquered.
